The GABA A receptor alpha 1 subunit (GABRA1) is a critical component of the GABA A receptor complex, a family of fast-acting ligand-gated ion channels that mediate the majority of rapid inhibitory neurotransmission in the central nervous system. These receptors are pentameric structures composed of various subunits, with the alpha 1 subunit being the most abundantly expressed in the adult brain. GABRA1 plays a central role in maintaining synaptic inhibition, regulating neuronal excitability, and shaping network oscillations essential for cognitive and behavioral functions. Emerging evidence links GABRA1 dysfunction to the pathophysiology of several neurodegenerative diseases, including Alzheimer’s disease, Parkinson’s disease, and Huntington’s disease. Altered expression or function of the alpha 1 subunit has been associated with key features of neurodegeneration, such as disrupted inhibitory signaling, synaptic imbalance, and increased neuronal vulnerability. As such, GABRA1 is gaining attention as a potential biomarker and therapeutic target in the early detection and treatment of these disorders. Advancements in molecular imaging, transcriptomics, and electrophysiology are enhancing our understanding of GABRA1’s spatial distribution and functional dynamics in both healthy and diseased brains.
