The canonical Wnt pathway controls cell fate, proliferation, and stem-cell maintenance, and its dysregulation is a driving force in many cancers, most notably colorectal carcinoma. The central event is the regulated stability of beta-Catenin. In the absence of Wnt ligand, a cytoplasmic destruction complex assembled around the scaffolds APC and Axin1 positions beta-Catenin for sequential phosphorylation; GSK-3beta, acting together with CK1, phosphorylates beta-Catenin to create the Phospho-beta-Catenin species recognized by an E3 ligase, leading to its ubiquitination and proteasomal degradation. This constitutive turnover keeps nuclear beta-Catenin low and target genes silent. Wnt ligand binding to Frizzled and LRP co-receptors recruits and activates the cytoplasmic protein Dvl (Dishevelled), which inhibits the destruction complex, in part through inhibitory modification of GSK-3beta detectable as Phospho-GSK-3beta. With the destruction complex disabled, beta-Catenin accumulates, escapes degradation, and translocates to the nucleus, where it partners with TCF/LEF transcription factors such as TCF4/TCF7L2 to activate genes that promote proliferation and stemness, including MYC and CCND1. In colorectal and other cancers, the pathway is constitutively activated most often by truncating mutations in APC, or alternatively by stabilizing mutations in beta-Catenin or loss of Axin1, all of which permit unchecked nuclear beta-Catenin accumulation. This central role has made the pathway an intensively pursued therapeutic target. Measuring total and phosphorylated beta-Catenin, the activity-linked phosphorylation of GSK-3beta, the levels of the scaffolds APC and Axin1, and the transcriptional partner TCF4/TCF7L2 together with Dvl provides a comprehensive view of pathway state.
