Myelination is the process by which oligodendrocytes in the central nervous system wrap axons in multilayered lipid-rich sheaths that enable rapid saltatory conduction and provide metabolic and trophic support to axons. Disruption of myelin underlies demyelinating diseases such as multiple sclerosis and contributes to the axonal degeneration that follows. The oligodendrocyte lineage is specified and maintained by transcription factors, and two are central to this program. Sox10 is a master regulator expressed throughout the lineage that is required for oligodendrocyte differentiation and the expression of myelin genes, while Olig2 establishes oligodendrocyte identity from progenitors and persists in mature cells. Oligodendrocyte precursor cells, the dividing pool that generates new myelinating cells during development and remyelination, are identified by NG2/CSPG4, a surface proteoglycan that marks this population. As progenitors mature they express the structural constituents of compact myelin. MBP, myelin basic protein, is a major component that compacts the cytoplasmic surfaces of the myelin membrane, and PLP1, proteolipid protein, is the most abundant integral myelin protein that stabilizes the sheath. CNPase is an early and abundant oligodendrocyte enzyme enriched in non-compact myelin and used as a lineage and myelination marker. Two glycoproteins reside in specialized myelin domains: MAG, myelin-associated glycoprotein, mediates axon-glia interactions at the periaxonal membrane and signals to maintain axonal integrity, and MOG, myelin oligodendrocyte glycoprotein, sits on the outermost surface of myelin and is a prominent autoimmune target in demyelinating disease. Together these markers track the full trajectory from precursor specification to mature myelin assembly.
