Cytokines are secreted signaling proteins that coordinate the initiation, amplification, and resolution of immune and inflammatory responses by acting on specific cell-surface receptors. The balance between proinflammatory and anti-inflammatory cytokines determines whether an immune response clears a threat and resolves or progresses to chronic inflammation and tissue damage. Tumor necrosis factor alpha (TNF-alpha) is a master proinflammatory cytokine that signals through TNFR1 to activate NF-kB and MAP kinase cascades, inducing inflammatory gene expression, and under certain conditions can trigger apoptosis or necroptosis.
