Neurotrophins are secreted growth factors that govern neuronal survival, differentiation, synaptic plasticity, and the formation and maintenance of circuits throughout life. They signal through two receptor classes: the high-affinity Trk receptor tyrosine kinases, which mediate trophic and survival responses, and the pan-neurotrophin receptor p75, which can promote either survival or apoptosis depending on context. The most abundant neurotrophin in the adult brain is BDNF, which is essential for activity-dependent synaptic plasticity, long-term potentiation, and learning and memory. BDNF binds its cognate receptor TrkB/NTRK2, triggering receptor dimerization and autophosphorylation; the activated, phosphorylated receptor, detected as Phospho-TrkB, recruits adaptor proteins that engage the Ras-ERK, PI3K-Akt, and PLC-gamma cascades to support neuronal survival and synaptic strengthening.
