Neuroligin 1 (NLGN1) is a postsynaptic cell adhesion molecule critical for the formation and maturation of excitatory synapses. As a member of the neuroligin family, NLGN1 interacts with presynaptic neurexins to establish trans-synaptic connections that regulate synaptic specificity and plasticity. In the adult brain, NLGN1 is essential for long-term potentiation (LTP), synaptic remodeling, and memory formation, particularly within the hippocampus and amygdala. Dysregulation of NLGN1 expression or function has been linked to impaired synaptic transmission, cognitive deficits, and neurodevelopmental disorders. In neurodegenerative diseases, such as Alzheimer’s disease, synaptic loss precedes neuronal death. Altered NLGN1 signaling may contribute to early synaptic dysfunction, making it a potential biomarker and therapeutic target.
