Immunofluorescence staining of Hela cells with PACO60032 at 1:133, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L). |
バックグラウンド: | Protein tyrosine phosphatase which may play a role in the regulation of lymphangiogenesis, cell-cell adhesion, cell-matrix adhesion, cell migration, cell growth and also regulates TGF-β gene expression, thereby modulating epithelial-mesenchymal transition. Mediates beta-catenin dephosphorylation at adhesion junctions. Acts as a negative regulator of the oncogenic property of YAP, a downstream target of the hippo pathway, in a cell density-dependent manner. May function as a tumor suppressor. |
シノニム: | Tyrosine-protein phosphatase non-receptor type 14 (EC 3.1.3.48) (Protein-tyrosine phosphatase pez), PTPN14, PEZ PTPD2 |
UniProt Protein Function: | PTPN14: a non-receptor phospho-tyrosine protein phosphatase. Reported to be mutated in a small percentage of human cancers including colorectal cancers and a smaller fraction of lung, breast, and gastric cancers. May play a role in liver metastases and tumour invasion in pancreatic cancer. Contains 1 FERM domain. |
UniProt Protein Details: | Protein type:Protein phosphatase, tyrosine (non-receptor); Motility/polarity/chemotaxis; EC 3.1.3.48 Chromosomal Location of Human Ortholog: 1q32.2 Cellular Component: cytoplasm; nucleoplasm; nucleus Molecular Function:protein binding; protein tyrosine phosphatase activity; receptor tyrosine kinase binding; transcription cofactor activity Biological Process: lymphangiogenesis; negative regulation of cell proliferation; protein amino acid dephosphorylation; regulation of protein export from nucleus Disease: Choanal Atresia And Lymphedema |
NCBI Summary: | The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP contains an N-terminal noncatalytic domain similar to that of band 4.1 superfamily cytoskeleton-associated proteins, which suggested the membrane or cytoskeleton localization of this protein. It appears to regulate lymphatic development in mammals, and a loss of function mutation has been found in a kindred with a lymphedema-choanal atresia. [provided by RefSeq, Sep 2010] |
